Mechanisms of Protein Biogenesis
Proteins are the main effectors of biological processes, and their synthesis requires the concerted action of sophisticated molecular machines within cells. Their lives begin on the ribosomes, which ensure the faithful translation of the genetic code by pairing each mRNA codon with the correct amino acid through a matching tRNA. Events during these formative minutes can have long-lasting consequences, since many nascent proteins start to fold and even interact with their binding partners as they emerge from translating ribosomes.Our research seeks to define how eukaryotes establish and maintain functional proteomes at the molecular, cellular, and organismal level, with the long-term goal of understanding why some cell types are particularly vulnerable to the dysregulation of these processes in disease. To address these questions in physiological settings, we develop and employ quantitative genome-wide approaches (ChIP-Seq, tRNA-Seq, ribosome profiling) and combine those with functional genomics in human pluripotent stem cell-derived models.
