Steady-state operation of a cell-free band-detection gene circuit


Due to the exhaustion of cell-free gene expression systems over time, it is difficult to realize cell-free gene circuits in closed reactors that require extended runtimes for operation. In this work we show that execution of an incoherent feed-forward loop of type 2 (IFFL-2)  - capable of distinguishing between high, intermediate and low input signals - can be successfully established in microfluidic ring reactors, which allow running the circuit under steady state conditions.
